Danon Disease Market Outlook and Forecast
Thelansis’s “Danon Disease Market Outlook, Epidemiology, Competitive Landscape, and Market Forecast Report – 2026 To 2036” covers disease overview, epidemiology, drug utilization, prescription share analysis, competitive landscape, clinical practice, regulatory landscape, patient share, market uptake, market forecast, and key market insights under the potential Danon Disease treatment modalities options for eight major markets (USA, Germany, France, Italy, Spain, UK, Japan, and China).
Danon Disease Overview
Danon disease is a rare X-linked lysosomal and autophagic disorder caused by pathogenic variants in the LAMP2 gene, resulting in impaired autophagic flux and accumulation of undegraded cellular material. The disease is characterized by hypertrophic cardiomyopathy, skeletal myopathy, conduction abnormalities, ventricular arrhythmias, and progressive heart failure, with neurologic and ophthalmologic manifestations also occurring. Males typically develop more severe and earlier-onset disease than females. There is currently no approved disease-specific pharmacotherapy, and management remains multidisciplinary, focusing on cardiac and extracardiac complications, arrhythmia control, and heart transplantation for selected patients with advanced cardiac disease. The therapeutic landscape is now expanding toward gene replacement, led by RP-A501, an AAV9-based therapy delivering the LAMP2B transgene. Phase 1 data demonstrated myocardial LAMP2 expression with encouraging cardiac findings over 24–54 months, while a single-arm Phase 2 study is evaluating the therapy in male patients with Danon disease. These results provide proof-of-concept for gene-based treatment, although durability, long-term clinical benefit, safety, and applicability across the broader patient population remain important areas of investigation.
